改正GMP省令の第3条の4 品質リスクマネジメントには以下の要求事項がある。
製造業者等は、品質リスクマネジメントを活用して医薬品品質システムを構築した上で、医薬品に係る製品について、製造所における製造管理及び品質管理を行わなければならない。

薬生監麻発0428第2号の逐条解説では下記のように説明されている。
６．第３条の４（品質リスクマネジメント）関係
（２）医薬品の製造業者等があらかじめ指定した者に行わせる品質リスクマネジメントの実施の手続きその他の必要な事項に係る文書及び記録の作成及び保管について規定するものであること。
あらかじめ指定した者については、品質リスクマネジメントに関して熟知している職員を当該文書及び記録の作成及び保管の責任者としてあらかじめ指定し、その職責及び権限を含め、GMP省令第６条第４項の規定による文書に適切に定めておくことが求められる。
ここで注意しなければならないことは、『品質リスクマネジメントに関して熟知している職員』といった要求である。
品質リスクマネジメントは、製造や試験検査における逸脱等により、医薬品等の品質に欠陥が生じた場合、患者にどういった健康被害（危害）が生じるかを推論するものである。
つまり、薬学・医学的な専門知識を必要とする。
果たしてそのような知識を持った職員が製造所（製造業者）に存在するかどうかである。
ICH Q9で要求され、今般改正GMP省令に盛り込まれた品質リスクマネジメントは、適正に実施することは非常に困難を伴う。
製造販売業者の開発担当者（技術移転のスタッフなど）と連携することが求められる。
外部委託業者管理責任者
改正GMP省令の第11条の5 第2項には以下の要求事項がある。
製造業者等は、あらかじめ指定した者に、手順書等に基づき、次に掲げる業務を適切に行わせなければならない。
一 外部委託業者との取決めの締結に際して、当該外部委託業者の適性および能力について確認すること。
二 外部委託業者が当該委託に係る製造・品質関連業務を適正かつ円滑に行っているかどうかについて定期的に確認するとともに、必要に応じて改善を求めること。
三 前2号の業務に係る記録を作成し、これを保管すること。

（２）医薬品の製造業者等があらかじめ指定した者に行わせる外部委託業者の管理に係る業務について規定するものであること。
