1.　生デ電子生データの留意点ータ信頼性向上の留意点
1.1 生データとは
製薬業界では、生データの取り扱いについては、周知されているものと思われる。
生データとは、最初に文字化または記号化されたものである。
万が一、検査の結果をシャーレのふたに記録したとすれば、当該記録（シャーレのふた）が生データとなる。
もちろん、そんなことをしてしまうと、後から大変なことになってしまう。
正しくは、記録を紙のシートに最初に記載し、それをシャーレのふたに転記するのである。
また、観察結果は速やかに記録されねばならない。
1.2 生データの変更箇所は要注意
誤記訂正など、記載を間違えた箇所には問題が潜んでいる可能性がある。
したがって、変更理由、変更箇所、変更前の数値、署名、日付けなどを入念に確認する
なぜならば、間違えたそれなりの理由があるはずだからである。
少し気が緩んでいたとか、勘違いしてしまったなどである。
しかしながら、勘違いしたと思ったことが勘違いであったという場合もある。
実は正しく書いているのに、間違ってしまったと思い込んでしまうなどである。
したがって、記録を訂正する場合には、最初の記載を見えるように二重線で消すことが基本である。
黒く塗りつぶしたりしてはならないのである。
こういうことは、すべての従業員に徹底して教育しておかなければならない。

2.　GLP における生データの定義
GLP における生データの定義を見てみたい。
21 CFR Part 58.3(k) には、以下の記載がある。
“ 生データ” とは、実験室内のワークシート記録書、覚え書き、注意書き、またはその正確なコピーをいい、これは非臨床試験の原観察結果およびその業務についての成績であり、この試験の報告の再構成および評価のために必要である。
生データの正確な転写( 例えば、そのまま転写され、日付をつけ、署名によって正確であると確認されたテープ) が用意された場合には、その正確なコピーまたは正確な転写を生データとしてもとの資料と置き換えることができる。
生データは写真、マイクロフィルムまたはマイクロフィッシュ、コンピュータ記録、観集結果を口述した磁気記録および自動装置から記録されたデータでもさしつかえない。
例えば、測定機器等から出力された感熱紙などは、時間とともに茶けてしまい、いずれ読み取れなくなってしまう。
そのような場合、コピーを取っておかなければならない。
コピーする際には、実施した者の署名と日付を記載しておくこと。
このような署名付きのコピーのことをCertifi ed Copy という。
Certifi ed Copy は、生データとして定義することができる。
また、21 CFR Part 58.130(e) には、以下の記載がある。
非臨床試験実施中に得られたデータは、自動化されたデータ収集システムにより得られたデータを除いて、すべて直接、直ちにかつ読みやすくインクで記録されなければならない。
すべてデータの記入に際しては、記入日付を記載し、データ記入者の署名または略署名がなければならない。
いかなる記入事項の変更も最初の記入事項を不明瞭にしないような方法で行ない、その変更の理由を示し、かつ、変更の時点で日付を記載し、署名するか、または変更者を明らかにしなければならない。
自動化されたデータ収集システムにおいては、データ入力時点でデータの直接入力責任者を明らかにしなければならない。
自動化されたデータの記載事項中のいかなる変更についても、原本の記載を不明瞭にしないような方法で行ない、その変更理由を示し、日付を記載し、責任者を明らかにしなければならない。
前半が、手書きによる生データの記録と訂正方法であり、後半が自動化された生データの記録と訂正方法である。
自動化された生データの記録方法には、種々の方法が存在する。
例えば、分析機器にSD カード等の不揮発性メモリーなどがついており、それらに記録する方法がある。またインテグレータやパソコンが接続されており、ハードディスクに記録されるもの。さらに、分析機器等からLIMS に接続されており、データベースに記録されるものもある。記録が紙であれ電子であれ、本質的には記録した者、日付（電子の場合には時刻）が必要であり、訂正する場合には、オリジナルが見えにくくならないように消して、新たな値等を記載し、訂正理由を記録しなければならない。電子の場合には、監査証跡がそれにあたる。
3.　電子生データの特徴
読者は、紙の生データと電子の生データは違いがないものと思っているかも知れない。
実はそうではないのである。
紙の生データの場合には、その定義は明らかである。
しかしながら、電子生データの場合には、様相が違ってくる。
なぜならば、電子は何回コピーしても” 生” だからである。
